There are 2 tarred roads to Vleesbaai from the highway (approx 15km drive).

On the N2, travelling from Cape Town towards Mosselbay
pass Albertinia
take the first turn-off to the right towards Gouritzmond
turn left towards Vleesbaai at the first crossing, 
cross the bridge and turn right at the T junction
turn right at the next tarred road towards Vleesbaai  

On the N2 travelling from George Airport towards Cape Town,
pass Mossel Bay
take the first turn-off to the left towards Vleesbaai
(opposite the Petro SA).
Pass the turn-offs to Nautilus Bay, Springer Bay and Boggomsbaai
Turn left at the next road towards Vleesbaai.

Go through the security entrance, turn left at the first tarred road at the “Driehoek” signboard (before the Vleesbaai Supermarket), and right at the next street.  We are at the end of the cul-de-sac)

NOTE:  In GoogleMaps, the street is mispelled as RYMA (correct - Rymar)

It is 80km away from George airport and in the centre of the Garden Route, halfway between Cape Town and Port Elizabeth (both 400km away)
